A worked example, step by step
Take a 10 mg vial of retatrutide. First convert the label to micrograms, because working amounts are usually discussed in micrograms: 10 mg is 10,000 mcg. Now add 2 mL of bacteriostatic water. The concentration is 10,000 mcg divided by 2 mL, which gives 5,000 mcg per mL, or 5 mg/mL. That number, micrograms per millilitre, is the anchor for everything that follows.
From concentration to units on the syringe
A standard insulin syringe is marked in units, where 100 units fill exactly 1 mL. So at 5,000 mcg/mL, one full millilitre (100 units) holds 5,000 mcg, and each single unit holds 50 mcg. To measure an example aliquot of, say, 500 mcg for a protocol, you divide 500 by 50 and draw to the 10-unit line. Why the solvent volume is the lever
Notice from the chart that the vial always contains the same 10,000 mcg. Reconstituting with 1 mL makes a solution four times more concentrated than reconstituting with 4 mL, and the units-per-aliquot change accordingly. A more concentrated solution means smaller, harder-to-read draws; a more dilute solution means larger, easier-to-read draws but a bigger fill volume. Many researchers pick a volume that puts their common aliquot at a round, readable number of units. Recording the exact volume you used is what makes any figure reproducible later.
